RNeasy PowerFecal Pro Kit (50)

An extraction kit from QIAGEN that isolates microbial RNA from stool and gut samples, sludge, or wastewater. The workflow begins by homogenizing each sample in a 2 ml bead beating tube containing a mixture of lysis beads, where mechanical collisions between beads combined with chemical disruption of cell membranes allow lysis of host and microbial cells. The lysate is then passed through an MB RNA Spin Column, DNA is removed using on-column DNase digestion, and the RNA is eluted with RNase-free water for immediate use in downstream applications.

QIAGEN's second-generation Inhibitor Removal Technology (IRT) removes substances commonly found in stool samples that inhibit PCR and other downstream applications, supporting high yields of highly pure RNA with minimum copurification of DNA. Sample types include stool, gut material, sludge, and wastewater, with a recommended starting input of 50-200 mg per sample (the manufacturer recommends starting with 50-100 mg of stool). The kit is supplied in a 2 ml tube format. For reagent storage, CD2 and lyophilized DNase I should be kept at 2-8°C, while all other kit reagents and components should be stored at room temperature (15-25°C).

Isolated RNA is compatible with immediate use in downstream applications including RT-PCR, qPCR, digital PCR and next-generation sequencing (e.g., RNA-seq or metatranscriptome). RNA isolation can be fully automated on the QIAcube Connect or the classic QIAcube; the QIAcube Connect processes up to 12 samples per run using spin column technology. Sample integrity ahead of extraction can be supported by the PowerProtect DNA/RNA reagent, which stabilizes stool samples at room temperature.
