RNeasy DSP Midi Kit

QIAGEN's RNeasy DSP Midi Kit is a spin-column kit designed for manual isolation and purification of total RNA from human blood and bone marrow aspirates, intended for use with in-vitro diagnostic QIAGEN products that list the RNeasy Midi Kit as an accessory. The kit employs silica-membrane technology (RNeasy technology), which combines the stringency of guanidine-isothiocyanate lysis with the speed and purity of silica-membrane purification.

Erythrocytes from blood are lysed (erythrocyte lysis buffer not included in the kit) and white blood cells are collected, lysed, and homogenized. Ethanol is added to provide ideal binding conditions, and the lysate is loaded onto the RNeasy Midi column. Subsequent washing and centrifugation steps are applied before RNA is eluted in RNase-free water at a volume of 300-500 µL. Processing is manual and performed by centrifugation.

RNA purified using the kit can be used directly for sensitive analytical detection in downstream assays such as qPCR, or further concentrated using the RNeasy DSP MinElute Cleanup Kit. Detailed protocols are provided in the instructions for use of the corresponding downstream applications.
