Integration-free reprogramming of somatic cells into human induced pluripotent stem cells (iPSCs) is achieved through a single vial kit supplied by STEMCELL Technologies, delivering modified synthetic mRNAs that encode OCT4, KLF4, SOX2, GLIS1, and c-MYC transcription factors. The five-factor OKSGM combination is administered across five consecutive daily transfections, paired with a recombinant B18R interferon inhibitor that suppresses the innate antiviral response normally triggered by exogenous RNA in target cells. Researchers typically begin with dermal fibroblasts or peripheral blood mononuclear cells maintained under feeder-free conditions, then transfer successfully reprogrammed colonies to TeSR medium within 7 to 10 days after the final transfection. Each kit ships as a single-use reagent set that includes the mRNA factor cocktail, the B18R supplement, and a transfection-grade additive, with all components stored at -80 °C and protected from light to preserve RNA stability. Because no viral vectors or genomic integration are required, the protocol yields footprint-free iPSC lines suited for downstream disease modeling, drug screening, and in vitro research use only applications, including generation of patient-specific lines for cell culture workflows.
