REPLI-g Mini Kit (100)

This QIAGEN whole genome amplification system is designed for use with purified genomic DNA, blood, and cells. Using Multiple Displacement Amplification (MDA) with Phi29 DNA polymerase, it performs isothermal amplification at 30°C with a uniquely processive DNA polymerase carrying 3'–5' exonuclease proofreading activity, providing highly uniform amplification across the genome with negligible sequence bias.

Typical yield is approximately 10 µg per 50 µL reaction, with an average product length greater than 10 kb and a range from 2 kb to 100 kb. The protocol is optimized for >10 ng of purified genomic DNA template; smaller inputs of 1–10 ng can be used when DNA quality permits. Reactions are incubated at 30°C for 10–16 hours, after which the polymerase is inactivated by heating at 65°C for 3 minutes. The amplified DNA is suitable for downstream applications including next-generation sequencing, array analysis, and genotyping.

Kit components include DNA polymerase, reaction buffer, Buffer DLB, Stop Solution, PBS, 1 M DTT, and a Quick-Start Protocol. The kit is shipped on dry ice and should be stored at −20°C upon receipt, where it remains stable for at least 6 months. It is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of disease.
