REPLI-g Midi Kit (100)

A high-yield whole genome amplification kit from QIAGEN designed for laboratories requiring amplified DNA from limited samples. The kit employs isothermal Multiple Displacement Amplification (MDA) chemistry using Phi29 DNA Polymerase, delivering fidelity reported as up to 1000 times that of Taq DNA Polymerase. The reaction supports DNA input across a size range of 2 kb to 100 kb, and from a starting input of 10 ng purified genomic DNA the midi-scale reaction yields up to 40 µg of amplified product.

The amplified DNA is of high molecular weight, making it suitable for demanding downstream workflows including next-generation sequencing (NGS), array analysis such as comparative genomic hybridization, long-range PCR, SNP genotyping with TaqMan primer/probe sets, qPCR- and PCR-based mutation detection, STR/microsatellite analysis, Sanger sequencing, RFLP and Southern blot analysis, and biomarker research. The reaction is optimized for larger reaction volumes to maximize yield in high-throughput settings and provides uniform genome coverage across the amplified product.

Designed as a bulk midi-scale kit for whole genome amplification, it operates as a manual laboratory procedure with hands-on time of approximately 20 minutes. Reagents should be stored at -20 °C and the recommended protocol followed for reproducible performance.
