Repli-g Advanced DNA Single Cell Kit(96)

This QIAGEN kit enables high-fidelity whole genome amplification from single cells or purified genomic DNA using Multiple Displacement Amplification (MDA) technology. The system is optimized for sensitive microarray and next-generation sequencing applications, providing high uniformity with minimal allelic dropout and negligible sequence bias. Buffers and reagents undergo a controlled decontamination procedure to minimize contaminating DNA. The kit should be stored immediately upon receipt at –30 to –15°C for up to 6 months, or at –90 to –65°C for longer storage.

Protocol 1 is designed for 1–1000 intact eukaryotic single cells and is not suitable for cells treated with formalin or other cross-linking agents. Protocol 2 is intended for 1–10 ng of purified genomic DNA suspended in TE buffer, with best results when template DNA is >2 kb in length with some fragments >10 kb. Typical DNA yields are 20–30 µg per 50 µl reaction. A supplementary protocol covers whole genome amplification from flash-frozen tissue sections, where yields are approximately 25–35 µg per 50 µl reaction.

Amplified DNA is suitable for downstream applications including next-generation sequencing, array CGH, and quantitative PCR. For PCR analysis, an aliquot of amplified DNA is diluted 1:100 and 2 µl of the diluted DNA is used per reaction. The kit is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of disease.
