Repli-g Advanced DNA Single Cell Kit(24)

A whole genome amplification kit from QIAGEN enables highly uniform amplification from single eukaryotic cells, limited sample amounts, or isolated genomic DNA. The kit uses Phi29 polymerase in a Multiple Displacement Amplification (MDA) reaction, offering 1000x greater accuracy than Taq-based methods, with advanced cell lysis and improved chemistry that increase uniformity and reduce amplification bias.

The simplified workflow comprises three steps with about 20 minutes of hands-on time: alkaline cell lysis for 10 minutes at room temperature, a 2-hour MDA reaction at 30 °C, and inactivation for 3 minutes at 65 °C. Typical yields are 25–35 µg of amplified DNA from a single cell; a single eukaryotic cell is estimated to contain 6 pg of DNA, an input insufficient for most genomic downstream applications. The kit is compatible with microarray, qPCR, and next-generation sequencing analysis, Sanger sequencing on capillary sequencers, and Pyrosequencing on PyroMark systems, and amplified DNA can be used in NGS, array CGH and quantitative PCR.

The kit should be stored at –30 to –15 °C upon receipt, and amplified DNA may be held at 4 °C for short-term use or at –20 °C long term at a recommended concentration of at least 100 ng/µl. The product is intended for molecular biology applications and is not for the diagnosis, prevention or treatment of disease.
