Description

An imidazoquinoline compound supplied by STEMCELL Technologies, R848 acts as an immune-response modifier and agonist of Toll-like receptors (TLRs) 7 and 8. It mimics pathogen-associated molecular patterns to activate immune cells, with reported anti-tumor and anti-viral activity. The compound demonstrates an IC₅₀ of 4.2 µM, with effects mediated predominantly through induction of cytokines such as interferon (IFN)-α and interleukin (IL)-12 following stimulation of monocytes, macrophages, and dendritic cells.

In immunology research, the compound triggers activation of human B cells, induces proliferation and cytokine production by human CD4+ T cells, primes human neutrophils for leukotriene B4, prostaglandin E2, and platelet-activating factor biosynthesis, suppresses HIV-1 replication in monocytes, and induces expression of IL-12 and IFN-γ in mouse and human peripheral blood cell cultures. In differentiation studies, it targets osteoclast precursors to inhibit their differentiation into osteoclasts via TLR7 and induces myeloid differentiation of CD34+ hematopoietic progenitor cells, upregulating cytokines including IL-1β, TNF-α, IL-6, and GM-CSF and the CD11c surface marker.

The product is supplied as a crystalline solid with a purity of ≥ 98%. Chemical properties include molecular formula C₁₇H₂₂N₄O₂, molecular weight 314.4 g/mol, and CAS Number 144875-48-9. Alternative names are Resiquimod and S 28463. The compound is stable at -20°C when supplied, should be protected from prolonged light exposure, and stored with a desiccant for long-term storage. R848 is soluble in DMSO (≤ 35 mM) and absolute ethanol (≤ 10 mM) but has low solubility in aqueous media; for cell culture use, stock solution should be diluted into culture medium immediately before use, keeping the final DMSO concentration below 0.1%.
