QuickLyse Miniprep Kit (250)

QIAGEN's plasmid DNA purification platform uses one-step lysis technology to rapidly isolate sequencing-grade plasmid DNA. The QuickLyse Miniprep Kit combines enzymatic and osmotic processes to lyse bacterial cells in a single 3-minute step, eliminating the need for a separate lysate-clearing centrifugation. Bacterial cell resuspension, lysis, and DNA binding are achieved in a single buffer, enabling preparation of 24 plasmid DNA minipreps in less than 22 minutes.

The kit delivers up to 15 µg of high-copy plasmid DNA from 1.5 ml of bacterial culture, with an elution volume of 50 µl. Throughput is 1–24 samples per run, and processing is manual using centrifugation at 13,000 rpm (approximately 17,000 × g). Isolated plasmid DNA exhibits OD 260/280 ratios of 1.7–1.9, and sequencing read lengths of over 700 bases are easily achieved. A 1.5 ml overnight culture of high-copy plasmids typically yields 3–8 µg of plasmid DNA. The kit is optimized for high-copy plasmids and is recommended for plasmids up to 9 kb in size; low-copy plasmids are not recommended.

Purified DNA is suitable for automated sequencing, PCR, restriction analysis, cloning, ligation, and transformation. The included Elution Buffer (Buffer QLE) contains 10 mM Tris-Cl and 0.1 mM EDTA (pH 8.5); its very low EDTA concentration does not inhibit enzymatic downstream reactions such as PCR and cycle sequencing. Growth in LB (Luria-Bertani) medium is strongly recommended to obtain the optimum combination of DNA quality, yield, and concentration.
