Description

Intended for molecular biology applications, this QIAGEN gravity-flow, anion-exchange tip enables isolation of pure, high-molecular-weight genomic DNA from whole blood, buffy coat, lymphocytes, cultured cells, tissues, mouse tails, yeast, Gram-negative bacteria, and some Gram-positive bacteria. The procedure relies on the selectivity of QIAGEN Anion-Exchange Resin, yielding DNA sized 20–150 kb with an average length of 50–100 kb that is free of contaminants such as RNA, protein, and metabolites and has A260/A280 ratios between 1.7 and 1.9. It avoids phenol and chloroform, requires no expensive equipment, and can be completed in less than 3 hours, with hands-on time of 20 minutes for bacteria, 30 minutes for blood and cultured cells, and 40 minutes for tissue and yeast.

The 20/G format has a maximum DNA-binding capacity of 20 µg. Recommended inputs and typical yields are 1 ml whole blood (15–20 µg), 5 × 10^6 HeLa cultured cells (15–20 µg), 15 mg liver tissue (15–20 µg), 1 ml S. cerevisiae (18–20 µg), 0.6–1.2 ml E. coli (16–20 µg), and 1.2–1.8 ml B. subtilis (16–20 µg). Purified DNA is suitable for next-generation sequencing, Sanger sequencing, RFLP techniques, screening of embryonic stem-cell clones and transgenic animals, Southern blotting, and PCR. Genomic-tips should be stored dry at 15–25°C and remain stable for at least 2 years. The product is not intended for the diagnosis, prevention, or treatment of a disease.
