QIAGEN Plasmid Maxi Kit (10)

QIAGEN Plasmid Maxi Kit is part of the QIAGEN Plasmid Kits family, providing gravity-flow anion-exchange tips for purification of transfection-grade plasmid DNA. Lysate clearing and isopropanol precipitation are achieved by centrifugation. The kit processes 100–500 ml culture volumes and is suited for high-copy, low-copy, and cosmid plasmid DNA, with manual (gravity-flow) processing of one sample per run using anion-exchange technology.

Up to 500 µg of high-copy plasmid DNA can be purified per run, with a typical run time of 160 min. Purified plasmid DNA is suitable for applications such as transfection, cloning, sequencing, and capillary sequencing. The system avoids toxic substances such as phenol, chloroform, ethidium bromide, and CsCl, and the anion-exchange resin is designed to deliver DNA purity equivalent or superior to that obtained by two successive rounds of CsCl gradient centrifugation.

The kit can be stored at room temperature (15–25°C) for up to 2 years. The Maxi protocol uses a QIAGEN-tip 500, with 10 ml Buffer QBT for equilibration, 2 x 30 ml Buffer QC for washing, and 15 ml Buffer QF for elution. Recommended LB culture volumes are 100 ml for high-copy plasmids and 500 ml for low-copy plasmids.
