A two-phase hot-start mechanism enables true room-temperature reaction set-up and provides at least two hours of room-temperature stability, preventing premature enzyme activity and artifact formation. The kit includes an internal control RNA that is reverse-transcribed into cDNA and co-amplified with the target, reporting instrument or chemistry failures, assay set-up errors, and the presence of inhibitors. Its duplex capability allows simultaneous detection of the internal control and the target RNA in a single reaction.

A visual pipetting control uses an inert blue dye in the master mix combined with a yellow template dilution buffer, turning the resulting solution green to confirm correct reaction set-up. Integrated genomic DNA reduction improves quantitation accuracy by reducing the work and costs associated with separate DNase digestion. The kit supports precise quantitation of low-copy RNA targets over a wide linear dynamic range on common real-time cyclers.
