QuantiNova Probe RT-PCR Kit (100)

From QIAGEN, this real-time RT-PCR kit is designed for highly sensitive, ultrafast, quantitative analysis using sequence-specific probes. It enables reliable quantification of RNA targets such as total RNA from eukaryotes and prokaryotes, poly(A)-RNA, and in vitro-transcribed RNA, and is compatible with dual-labeled probes including TaqMan probes.

The system employs a novel two-phase hot-start mechanism. HotStaRT-Script Reverse Transcriptase is associated with an RT-blocker that keeps the enzyme nearly inactive at ambient temperature, permitting room-temperature reaction setup for up to two hours without loss of performance. Once the protocol begins with an RT step at 45°C, the reverse transcriptase is activated and cDNA synthesis is initiated, while DNase in the RT mix reduces contaminating genomic DNA by approximately 90%. The second phase relies on QuantiNova DNA Polymerase, kept inactive by QuantiNova Antibody and QuantiNova Guard until a 5-minute, 95°C incubation step activates it. The optimized buffer system includes Q-Bond, an additive that enables short cycling steps without sacrificing PCR sensitivity or efficiency.

Kit contents include 2x QuantiNova Probe RT-PCR Master Mix (containing QuantiNova DNA Polymerase, QuantiNova Probe RT-PCR Buffer, and dNTP mix), 100x QuantiNova RT Mix (with HotStaRT-Script Reverse Transcriptase, RNase Inhibitor, and DNase), QuantiNova Yellow Template Dilution Buffer, QuantiNova Internal Control RNA for monitoring successful reverse transcription, QuantiNova ROX Reference Dye for instruments requiring ROX, and RNase-Free Water. A built-in visual pipetting control confirms correct template addition. The kit is optimized for use with any real-time cycler and is shipped on dry ice, with storage at –15 to –30°C protected from light.
