A real-time (RT-) PCR detection solution from QIAGEN designed for sensitive, rapid identification of pathogen nucleic acids using sequence-specific probes. The kit supports 4-plex real-time detection of user-defined pathogen targets — including viruses, bacteria, and fungi — alongside the QuantiNova Internal Control (IC) RNA or DNA in a single reaction. Both the IC and assay reagents can be used as extraction or amplification controls, supporting correct interpretation of negative results.

The ready-to-use, ultrafast formulation enables convenient room-temperature reaction setup suitable for manual or automated workflows, with a built-in visual pipetting control that indicates correct reagent mixing. The kit is robust in the presence of inhibitors and accommodates degenerated primers and probes. Real-time PCR and one-step RT-PCR can be performed simultaneously using the same protocol, and combined amplification of pathogen targets with the IC increases in-process safety while minimizing handling errors.

A two-phase hot-start mechanism uses heat-mediated activation of the HotStaRT-Script reverse transcriptase at 50°C and the QuantiNova DNA Polymerase at 95°C, with the QuantiNova Antibody and QuantiNova Guard additive stabilising the inactive polymerase complex. The reaction setup remains stable for at least one hour at room temperature. The master mix includes an inert blue dye, while the QuantiNova Yellow Template Dilution Buffer contains an inert yellow dye; mixing the template with the buffer turns the master mix from blue to green, providing visual confirmation of correct pipetting. The buffer system combines K+ and NH4+ ions with the synthetic Factor MP and Q-Bond additive to support stable annealing and rapid cycling. The included QuantiTect Nucleic Acid Dilution Buffer stabilises RNA and DNA standards and prevents loss on plastic surfaces. The master mix can be stored at 2–8°C for up to 12 months.
