QuantiNova Multiplex RT-PCR Kit (500)

Designed for sensitive, probe-based multiplex real-time RT-PCR, this QIAGEN kit enables simultaneous detection of up to 5 targets per well, allowing reliable quantification of multiple RNA targets in a single reaction. The ready-to-use master mix formulation supports direct amplification from a single cell or up to 800 ng template for both low- and high-abundance targets. Its room temperature setup streamlines 1-step RT-qPCR and enables automation.

The kit employs a novel antibody-mediated hot-start mechanism combined with a QuantiNova Guard molecule, allowing reactivation of Taq polymerase in 2–3 minutes for increased specificity. It includes a newly developed RNase H-deficient Moloney Murine Leukemia Virus Reverse Transcriptase (M-MLV RT). A 2-phase hot-start procedure enables reaction setup at room temperature while preventing premature enzyme activity. This advanced kit family replaces the predecessor QuantiFast and Rotor-Gene product lines, offering improved handling, speed, and in-process safety tools.

An optional Internal Control (IC) RNA can be incorporated to monitor successful reverse transcription and detect inhibitors carried over from RNA extraction. The IC RNA is detected as a 200 bp amplicon, and a Cq difference greater than 2 between control and template-containing samples indicates potential inhibition or sample failure. Tracking dyes in the kit allow visual confirmation of manual or automated pipetting. ROX can be added before use to accommodate instruments such as ABI cyclers that require a reference dye, with different amounts suited for Low ROX or High ROX instruments.
