From QIAGEN, this custom LNA-enhanced, probe-based qPCR assay provides accurate and sensitive gene expression analysis of any RNA target using hydrolysis probe-based detection. The design supports the full Ensembl database for human, mouse and rat targets and is well-suited for situations where no predesigned assay is available, for discriminating splice variants and for verifying novel mRNAs or lncRNAs. Predesigned assays, pathway-focused panels and custom panels can be used together in the same experimental setup.

Built on QuantiNova chemistry, the workflow can be completed in under 2 hours and is robust enough to support room-temperature setup, automation and delayed run of assays. The high binding affinity of LNA bases increases the flexibility of primer and probe placement on the transcript, enabling better target discrimination and reliable quantification even for AU-rich targets, low-abundance transcripts, targets with high secondary structure and highly complex samples. High sensitivity delivers excellent amplification efficiencies down to 1 RNA molecule, making low-abundance targets such as lncRNAs easier to detect from less starting material. Increased specificity from LNA placement provides a high signal-to-noise ratio, allowing discrimination of sequences that differ by only a single nucleotide and eliminating non-specific amplification and primer-dimer formation. The FAM-labelled probe delivers high specificity and improved mismatch discrimination.

Designed with a sophisticated algorithm incorporating over 50 different lab-validated parameters and used to design over 1.3 million assays, the custom assay builder in GeneGlobe accepts targets of at least 55 nucleotides and lets you submit up to 10 different target sequences at once.
