Description

QIAGEN's reference LNA probe assay uses a FAM-labelled reference probe to support consistent normalization and assay quality control in real-time PCR workflows. LNA modifications enhance the specificity and stability of probe binding, and the reference assay is designed for highly sensitive and rapid quantification of mRNA and lncRNA targets. The assay is compatible with QuantiNova master mixes and common real-time PCR platforms and, as part of the QuantiNova LNA Probe PCR System, works alongside the QuantiNova Reverse Transcription Kit and QuantiNova Probe PCR Kit to support accurate gene expression profiling for molecular biology applications. The product is for research use only and is not intended for the diagnosis, prevention, or treatment of a disease.

LNA (locked nucleic acid) oligonucleotides exhibit high thermal stability when hybridized to a complementary DNA or RNA strand. For each incorporated LNA monomer, the melting temperature (Tm) of the duplex increases by 2–8°C. As a result, LNA-based primers and probes can be made shorter than traditional DNA or RNA oligonucleotides while still retaining a high Tm, which is particularly advantageous when designing assays for AT-rich transcripts or when targeting specific isoforms and SNPs. Intelligent placement of LNA within primers can widen the Tm difference between perfect-match and mismatch targets, enabling better discrimination between closely related sequences, even those differing by only a single nucleotide.

Each lot of QuantiNova LNA Probe PCR Assays and Panels is tested against predetermined specifications under QIAGEN's ISO-certified Quality Management System to ensure consistent product quality. The QuantiNova DNA Polymerase used in the system is a modified form of a recombinant 94 kDa DNA polymerase originally isolated from Thermus aquaticus; it is provided in an inactive state with no enzymatic activity at ambient temperature and is activated by a 2 min 95°C incubation step. The QuantiNova LNA Probe PCR Assay portfolio includes over 1.3 million pre-designed assays, covering most transcripts of human, mouse, and rat genes in the Ensembl database. For two-step RT-PCR procedures, use with the QuantiNova Reverse Transcription Kit and the QuantiNova Probe PCR Kit is recommended to obtain optimal results.
