Description

Designed by QIAGEN for highly robust gene expression analysis, these probe-based real-time PCR assays use hydrolysis probe detection with FAM-labelled probes and locked nucleic acid (LNA) chemistry. LNA-modified primers and probes are designed shorter for greater positioning flexibility on the target, which increases specificity and mismatch discrimination. The combination of Tm-normalization and LNA enhancement gives a higher binding affinity, dramatically increasing assay sensitivity and specificity, with reported amplification efficiencies down to 1 RNA molecule. This enables sensitive detection of low-abundance targets such as lncRNAs from less starting material, while a high signal-to-noise ratio allows discrimination of sequences that differ by only a single nucleotide, eliminating non-specific amplification and primer-dimer formation.

Predesigned assays cover most transcripts in the Ensembl database for human, mouse and rat mRNAs and lncRNAs, with most assays intron-spanning where possible so they detect only RNA. Stringent design criteria and wet-lab validation are applied to popular mRNA and lncRNA targets, with up to three assay designs selected per transcript by an algorithm that incorporates over 50 different parameters. The QuantiNova hot-start mechanism enables successful qPCR even with difficult templates such as those of high GC-content or complex secondary structure, and is suitable for lncRNA analysis. The workflow can be completed in under 2 hours, supports room-temperature setup, automated setup and delayed run of assays, and the protocol has been tested on major brand qPCR cyclers. Assays are shipped on dry ice and in-stock items are typically delivered within 1–5 days. For research use only; not for diagnostic procedures.
