Description

LNA-enhanced predesigned assays from QIAGEN enable accurate and sensitive gene expression analysis of human, mouse and rat mRNA and lncRNA by qPCR or digital PCR. Locked nucleic acid (LNA) technology allows shorter primer design with flexible positioning on the target, and Tm-normalization provides higher binding affinity than standard DNA primers, dramatically increasing assay sensitivity and specificity. The primers discriminate sequences that differ by only a single nucleotide, eliminating non-specific amplification and primer-dimer formation, and deliver excellent amplification efficiencies down to 1 RNA molecule.

The assays have been developed using stringent design criteria and lab-validated algorithms incorporating over 50 parameters, with up to three designs per transcript fully wet-lab validated for popular mRNA and lncRNA targets. Predesigned assays cover most transcripts in the Ensembl database for human, mouse and rat genes; most assays are intron-spanning and detect only RNA. For SYBR Green-based qPCR, the workflow can be completed in 2 hours and supports room-temperature setup, automation and delayed run of assays. For digital PCR, the assays are validated on the QIAcuity instrument together with the QIAcuity EG PCR Kit using EvaGreen-based detection, providing highly precise target quantification.

Three assay categories are offered: gene-covering assays for broad transcript screening, transcript-specific assays optimized to detect a specific transcript, and splice variant transcript-specific assays for differentiating isoforms. Functionally validated reference gene assays are also available for data normalization. The assays are shipped at ambient temperature, and in-stock items are delivered within 1–5 days. Reverse transcription is performed using the QuantiTect Reverse Transcription Kit.
