Description

QIAGEN supplies this probe-based detection reagent as a 10x formulation for monitoring successful reverse transcription and amplification. The QuantiNova IC Probe Assay Red 650 detects the QuantiNova Internal Control RNA (QN IC RNA), which appears as a 200 bp amplicon, in the Cy5 or red dye channel of real-time PCR instruments. The assay carries a Cy5 analog label and is used together with the QuantiNova Probe PCR and RT-PCR Kits.

The assay is intended to report instrument or chemistry failures, errors in assay setup, and the presence of inhibitors such as phenol, ethanol, sodium dodecyl sulfate (SDS), or ethylene diaminetetraacetic acid (EDTA) that may remain from RNA lysis and purification. Its primer and probe sequences have been bioinformatically validated for non-homology against hundreds of eukaryotic and prokaryotic organisms, and experimentally tested against human, mouse, and rat RNA samples from multiple tissues and cell lines.

When used with the QuantiNova Probe or Multiplex RT-PCR Kits, the appropriate volume of the 10x assay is added to the reaction for signal detection in the Cy5 or red channel. A Cq difference greater than 2 between QN IC RNA-only controls and samples containing QN IC RNA plus template RNA is likely to indicate inhibition or sample failure. In that case, users are advised to check equipment for accurate performance, dilute the affected template RNA in RNase-free water, or repeat the RNA extraction using an appropriate RNeasy Kit or the RNeasy MinElute Cleanup Kit.
