Description
QIAGEN provides the Qproteome Nuclear Protein Kit for the specific enrichment of nuclear proteins from cultured mammalian cells. The workflow uses lysis and centrifugation to separate the cytosolic fraction (supernatant) from the cell nuclei (pellet). A high-salt buffer dissociates nuclear binding proteins, such as transcription factors, and removes them by diffusion from the nuclei.
Three fractions are isolated, with a binding capacity/yield of 200–300 µg from a starting sample of 5 × 10^6 cells in cell culture. The resulting nucleic acid binding protein fraction is suitable for downstream applications including SDS-PAGE and mass spectrometry, as well as a wide range of activity assays.
The kit is compatible with tissue samples using a supplementary protocol that employs the TissueRuptor.
