Qproteome Mitochondria Isolation Kit

From QIAGEN, this kit is designed for efficient isolation of high-purity mitochondria preparations using a fast, reproducible, and standardized procedure with a standard bench-top centrifuge. The workflow delivers fully active mitochondrial preparations in which proteins retain their native conformation and full biological activity, making the fractions well suited to apoptosis studies, 2D-PAGE analysis, and other biochemical assays.

Specifications: applications include SDS-PAGE, mass spectrometry, and activity assays; sample size ranges from 5 × 10⁶ to 2 × 10⁷ cells or 20–60 mg of starting material; binding capacity/yield is 20–80 µg of protein; species is Eukaryotes; one fraction is produced; start material is cells or tissue. Analysis of cell-compartment marker proteins indicates that the resulting mitochondrial fractions are virtually free of cross-contamination, which greatly facilitates analysis of low-abundance species.

The kit has been developed for cultured mammalian cells and has been tested with HeLa, Cos7, NIH, HEK 293, and SH-SY5Y cell lines. Animal tissue has not been tested, but may work if a cell-disruption method that leaves mitochondria intact is available. The kit is compatible with downstream reagents for protein isolation from the mitochondrial fraction and is also listed as compatible with tissue-sample protocols when used with a TissueRuptor.
