QIAwave RNA Plus Mini Kit (250)

A QIAGEN spin-column kit designed for purification of total RNA from animal cells and easy-to-lyse animal tissues. The procedure integrates QIAGEN's patented technology for selective removal of double-stranded DNA with well-established RNeasy technology, using a gDNA Eliminator column to clear genomic DNA contamination without the need for additional DNase digestion. Samples are lysed and homogenized in a highly denaturing guanidine-isothiocyanate-containing buffer that immediately inactivates RNases, then passed through the gDNA Eliminator before total RNA binds to the RNeasy membrane and is washed and eluted.

The kit is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of a disease. Purified RNA is ready to use and is suited to sensitive downstream applications, including quantitative real-time RT-PCR, next-generation sequencing, RT-PCR and digital PCR, differential display, cDNA synthesis, Northern, dot, and slot blot analyses, primer extension, Poly A+ RNA selection, RNase/S1 nuclease protection, and microarrays. The procedure isolates RNA molecules longer than 200 nucleotides, giving an enrichment for mRNA because RNAs shorter than 200 nucleotides, such as 5.8S rRNA, 5S rRNA, and tRNAs (which comprise 15–20% of total RNA), are selectively excluded. RNA is eluted in ≥30 µL of water.

Multiple samples can be processed in parallel in less than 25 minutes, replacing time-consuming methods such as CsCl step-gradient ultracentrifugation and alcohol precipitation, and avoiding the use of phenol or chloroform. The kit can be used manually or adapted for fully automated purification on QIAcube Connect or the classic QIAcube. Kits should be stored dry at room temperature (15–25°C) and remain stable for at least 9 months under these conditions.
