Description

From QIAGEN's QIAseq xHYB Human Hybrid Capture Panels portfolio, this panel is designed for hybridization capture-based enrichment of human exonic sequences from indexed whole genome NGS libraries for Illumina instruments. The QIAseq xHYB Actionable Exome Panel targets regions associated with known relevant genomic variants, with content built by leveraging variant information from the Human Gene Mutation Database (HGMD) and delivering high coverage of regions in over 10,000 genes. The panel size is 12.3 Mb.

Optimized probe design provides efficient coverage of challenging genomic regions, including high GC-content regions. Library fragments containing exonic sequences are specifically captured by biotinylated probes in as little as 30 minutes of hybridization, even from FFPE and cfDNA samples. The panel delivers 99% base-level coverage at ≥20x depth, enabling >98% combined sensitivity for SNVs and indels while minimizing dropouts. The single-day, automation-compatible sample-to-sequencing workflow is >33% faster than other exome workflows, and the QIAseq xHYB portfolio can reduce sequencing costs by up to 50% while providing high-quality SNV, indel and CNV calls.

The flexible workflow allows simultaneous hybridization capture from up to 8 samples with as little as 200 ng input per library. Pre-capture libraries are prepared with UDI-enabled QIAseq FX library preparation reagents, generating up to 384 unique dual-indexed libraries with a 2.5 hour workflow. The panel is also compatible with QIAseq cfDNA Library Kits and QIAseq Ultralow Input Library Kits. After capture on streptavidin-coated magnetic beads, exome-enriched libraries are amplified and prepared for sequencing on Illumina systems. Resulting FASTQ files are processed within the QIAGEN CLC Genomics Workbench for filtering, read mapping, and variant calling, with VCF output uploaded into QIAGEN Clinical Insight for variant filtering and prioritization. The product is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of a disease.
