Description

From QIAGEN, this RNA library preparation kit enables single-day, high-throughput next-generation sequencing (NGS) RNA-seq library construction for Illumina NGS instruments. It supports whole transcriptome or 3' RNA-seq workflows starting with total RNA or enriched mRNA, using ultraplex (UPX) cDNA pooling. The HMR configuration targets human, mouse and rat samples, depleting abundant host rRNA and other high-abundance transcripts while retaining coding and noncoding RNAs.

The workflow integrates QIAseq FastSelect for rapid ribosomal RNA removal during fragmentation and cDNA synthesis. In one step, QIAseq FastSelect removes up to 99% of all unwanted rRNA, even from degraded inputs such as formalin-fixed paraffin embedded (FFPE) samples. During reverse transcription, each transcript incorporates a unique sample ID. Following this step, 8 to 24 samples of cDNA are pooled (ultraplexed) and all subsequent library construction steps are performed in a single tube. During sample indexing and final library amplification, up to 768 different unique dual indices can be utilized, and when using the standard protocol, up to 18,432 samples can be sequenced together.

The kit supports library construction for transcript identification, fusion gene discovery and high-throughput gene expression studies from tissues, exosomes, FFPE samples and cell lines, with input RNA from eukaryotic, prokaryotic and fungal sources. The complete solution includes online access to an RNA-seq analysis portal for read alignment, differential gene expression and pathway analysis, and is also supported through QIAGEN's CLC Genomics Workbench.
