Description

This QIAGEN kit enables high-throughput 3' transcriptome analysis using next-generation sequencing on Illumina instruments. It supports library construction and gene expression analysis from purified RNA (10 pg – 10 ng), cell lysates (up to 1000 cells), and single cells. During reverse transcription, each cell is tagged with a unique Cell-ID and each RNA molecule with a unique molecular index (UMI), so cDNAs from each sample can be pooled and processed together in a single tube. Up to 48 different sample IDs can be used during final library amplification, and the combination of cell IDs and sample IDs allows up to 18,432 libraries to be sequenced together when starting with a 384-well plate.

The chemistry is LNA-enhanced for accuracy, specificity, and sensitivity, and UMIs eliminate library amplification bias to support accurate gene expression measurement. Data analysis is offered through cloud-based pipelines via QIAGEN GeneGlobe and through on-site software with QIAGEN CLC Genomics Workbench, enabling primary mapping, single-cell clustering analysis, and differential expression analysis. The product is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of a disease.

The kit is shipped in two boxes: Box 1 on dry ice (store at −30 to −15°C) and Box 2 on blue ice (store at 2–8°C). QIAseq Beads should never be frozen. When stored correctly, the kit is stable until the expiration date printed on the kit box lid, and each lot is tested against predetermined specifications under QIAGEN's ISO-certified Quality Management System. For blood-derived samples, QIAseq FastSelect –rRNA Globin can be used to remove globin mRNA.
