Single-cell transcriptomics library preparation reagent set from QIAGEN, configured for 96 reactions and supplied as the second-box indexing and amplification module of the multi-component QIAseq UPX 3' Transcriptome system. The chemistry supports ultra-high-throughput 3' mRNA profiling of mammalian cells using combinatorial indexing, allowing pool-of-cells processing that scales from several thousand to more than one million transcriptomes per sequencing run without traditional microfluidic encapsulation or droplet partitioning hardware.

Box 2 provides the indexed library-amplification reagents applied after upstream permeabilization and cell-barcode steps, enabling assembly of Illumina-ready libraries that read gene expression at the 3' end of polyadenylated transcripts. The workflow tolerates cryopreserved inputs, fixed tissues, and dissociated organoid or cultured-line samples, making it useful for cell atlasing, tumor heterogeneity studies, rare-population discovery, and CRISPR perturbation screens where batch integrity must be preserved across many wells.

Reagents are shipped for storage at -20 degrees C in a single-use kit format, with reaction volumes measured in microliters and indexed libraries compatible with standard Illumina sequencers. Researchers performing whole-transcriptome profiling can pair this box with the matching Box 1 components to generate sequencing-ready pools suitable for downstream single-cell bioinformatics analysis.
