Description

From QIAGEN, this kit is designed for preparing DNA libraries for next-generation sequencing (NGS) applications, producing Illumina-compatible libraries optimized for sensitivity and uniformity from limited starting material. It incorporates ultra-efficient end-repair and ligation chemistries into a streamlined solution for generating high-quality libraries from limited and damaged DNA sample types. The workflow completes in 2.5 hours and supports inputs from 10 pg to 100 ng of fragmented DNA.

Optimized enzymes and buffer formulations maximize performance with sub-nanogram samples while also accommodating up to 100 ng of input DNA. Library conversion rates exceed 30% for samples below 1 ng of input, with consistent conversion rates covering a 1000-fold range from 10 pg to 10 ng. A low percentage of adapter dimers helps minimize library contamination relative to the competition. The kit is suited for low-input whole genome sequencing, cfDNA for liquid biopsy, and limited sources of DNA from FFPE tissue, laser capture microdissection (LCM), and fine needle aspirations (FNA).

The protocol begins with double-stranded DNA fragmented enzymatically, chemically, mechanically or naturally, and uses an optimized end-polishing reaction with an Ultralow Input Ligation formulation alongside QIAGEN's proprietary HiFi PCR Master Mix. The same kit supports PCR-free library preparations from as little as 10 ng of DNA input, while PCR-based amplification is normally required below 100 ng. Dual-barcoded, plate-format adapters are included, with each well combining two eight-nucleotide identification barcodes (one i5 and one i7) per single-use adapter for multiplexed pooling. Cleanup uses Agencourt AMPure XP beads, and the streamlined protocol enables automation on various liquid-handling platforms.
