Targeted RNA sequencing offers a focused approach to transcript profiling, and QIAGEN's QIAseq Targeted RNAscan Extended Panel delivers expanded gene coverage for pathway-focused research. This NGS panel enables highly multiplexed analysis of curated gene sets on Illumina sequencing platforms, supporting studies in oncology, immunology, and cellular biology where deep, quantitative expression data on selected targets is more informative than whole-transcriptome sequencing.

The panel uses unique molecular indices (UMIs) integrated during reverse transcription and target amplification, minimizing PCR bias and enabling accurate transcript counting even from limited or degraded input. Libraries can be prepared from low-input total RNA samples, with each reaction processed in a 50 uL volume using provided primer pools. The extended gene content covers signaling pathways, immune markers, and differentiation-related transcripts beyond the scope of the standard RNAscan panel.

Workflow integration includes reverse transcription, targeted amplification, and indexing in a tube-based process that consolidates hands-on steps. Constructed libraries are stable when stored at -20 degrees C and are compatible with Illumina sequencers configured for 150 bp paired-end sequencing. Final reads are analyzed using GeneGlobe or related QIAGEN pipelines to generate count data normalized for UMI depth.

Typical applications include validation of RNA-seq discovery hits, biomarker screening in clinical research samples, pathway-level expression profiling, and characterization of FFPE-derived material where input quantity is limited. Each kit ships with all buffers, enzymes, and primer pools required for the complete library prep workflow, supporting up to 96 samples per package.
