QIAseq Targeted RNA Custom Panel (12) - 0 - 50 genes

QIAGEN's QIAseq Targeted RNA Custom Panels provide a Sample to Insight solution for quantitative gene expression profiling using RNA-seq. These panels integrate unique molecular indices (UMIs) with a two-stage PCR-based library preparation to deliver unbiased and accurate digital quantification from human, mouse and rat samples.

The workflow begins with converting total RNA into cDNA, with a typical RNA input of 25 ng of total RNA and no need for enrichment or rRNA depletion steps. The molecular indexing step uses a gene-specific primer (GSP1) carrying a 12-base UMI within a multiplex primer panel targeting 12–1000 genes. Following bead purification, a PCR enrichment is performed with GSP2 and RS2 primers, then a universal PCR with RS2 and FS2 primers adds sample-indexing barcodes to each sample.

Built-in control assays include gDNA assays to check for genomic DNA contamination and housekeeping gene (HKG) assays for data normalisation. QIAseq Targeted RNA Indices are available for indexing samples for targeted RNA sequencing. In a validation study using a 917-plex gene panel with 10 ng of NA12878 reference DNA, 97.5% of assays were within 20% of mean/median molecular tag counts.
