Description

QIAGEN's targeted RNA sequencing portfolio includes the QIAseq Targeted RNA 12-Index I, which provides sample-indexing barcodes and primers for targeted RNA sequencing on Illumina platforms. The index set is used alongside QIAseq Targeted RNA Panels during library preparation, where the indexing primers are added at the universal PCR step to add sample-indexing barcodes to each library and enable multiplex sequencing. The workflow requires minimal RNA input — as little as 25 ng of total RNA can be used — and no enrichment or rRNA depletion steps are necessary.

The panels use a digital sequencing approach in which a unique 12-base random molecular barcode is incorporated into gene-specific primers (GSP1) at the first extension step, immediately after mRNA is converted to cDNA. Because every original molecule is tagged before amplification, PCR duplicates and amplification bias can be removed at the analysis stage, yielding more accurate, unbiased gene expression quantification from human, mouse, and rat samples.

Built-in control assays include gDNA assays that detect any genomic DNA contamination in the RNA sample and housekeeping gene (HKG) assays for data normalization, supporting reproducible comparisons across samples and runs. Purchase of the panels also provides access to QIAGEN's GeneGlobe portal, which offers read alignment, data normalization, and differential gene expression analysis tools at no additional cost. The panels are compatible with Illumina MiSeq, NextSeq 500, HiSeq 1000, HiSeq 1500, HiSeq 2000, HiSeq 2500, and GAIIx sequencers.
