QIAseq Targeted Methyl Panel 8

QIAGEN's targeted methylation sequencing solution delivers a single-day workflow for interrogating DNA methylation status across thousands of CpG sites. The panel-based approach uses next-generation sequencing with integrated Unique Molecular Indices (UMIs) to enable sensitive and specific detection of methylation patterns from cells, tissues and biofluids.

The required amount of template for a single sequencing reaction ranges from 1–100 ng for fresh gDNA, 10–200 ng for FFPE DNA, or 10–100 ng for ccfDNA. The workflow begins with bisulfite conversion of isolated DNA (recommended: EpiTect Fast Bisulfite Kit), followed by DNA end repair. During target enrichment, UMIs are added to the library, allowing bioinformatics analysis to exclude duplicates and error-correct the sample's methylation status.

Built on QIAseq enrichment technology, the panels use a PCR method that pairs a single target-specific primer with a universal primer—an approach designed to maximise coverage of regions of interest while increasing sensitivity for challenging samples. The panels are described as delivering high mapping efficiency, high reproducibility and high correlation with established methods, and are compatible with difficult sample types including FFPE and liquid biopsy (ccfDNA).
