The Index Set D provides 96 unique dual-index adapters for multiplexing up to 96 individual bisulfite-converted DNA libraries in a single Illumina sequencing run. Part of the QIAseq Targeted Methyl workflow, these indices work alongside the panel's probe-based hybridization chemistry to enable quantitative, single-base resolution methylation profiling across user-selected genomic regions.

Set D is one of four complementary index sets (A through D) offered by QIAGEN, allowing laboratories to combine multiple sets and scale throughput up to 384 uniquely tagged samples per sequencing run. Each index contains an i7 and i5 sequence that supports demultiplexing on standard Illumina platforms, including NextSeq, MiSeq, NovaSeq, and HiSeq systems. The unique molecular index sequences also enable downstream deduplication and accurate quantification of methylation calls at low DNA input.

Researchers performing targeted methylation studies, including cancer epigenetics, imprinting disorder analysis, and cell-type-specific epigenetic surveys, use this index set to barcode their QIAseq Targeted Methyl libraries before pooling. The kit ships in a single-use plate format, with each well containing a single, pre-aliquoted index primer pair ready for direct addition during library construction.
