The workflow incorporates unique molecular indices (UMIs) into a single primer-based targeted enrichment process, facilitating ultrasensitive variant detection from cells, tissue, and biofluids while suppressing errors that limit detection of low-frequency variants in heterogeneous samples. The required template for a single sequencing reaction ranges from 10–80 ng for fresh DNA or 100–250 ng for FFPE DNA. Bead cleanups after ligation and target enrichment are replaced with enzymatic cleanups, delivering a streamlined workflow completed within 6 hours that is automation-friendly. An integrated FFPE DNA repair step prior to library construction improves recovery from formalin-fixed paraffin-embedded DNA samples.

The kit is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of disease. Quality is assured under QIAGEN's ISO-certified Quality Management System, with each lot tested against predetermined specifications. Compared to standard QIAGEN panels, these panels require 68% fewer pipette tips and use fewer reagents, with two separate index plates consolidated into a single plate containing both unique dual indexes (UDIs) in the same well. Shelf life has been extended from 7 months in the standard panels to 12 months.
