Description

This QIAGEN solution enables faster, automation-ready targeted next-generation sequencing (NGS) of DNA, with expanded variant analysis built on a digital sequencing foundation. Integrated unique molecular indices (UMIs) support confident detection of low-frequency variants including SNVs, small indels, CNVs, select structural variants, and loss of heterozygosity (LOH). Built-in FFPE DNA repair and optimized chemistry address fragmented DNA and GC-rich targets, making the QIAseq Targeted DNA Pro Panels suitable for DNA from tissue, blood, FFPE, and cfDNA.

Required input ranges from 10–80 ng of fresh DNA or cfDNA, or 100–250 ng of FFPE DNA, with a variable sample input volume of 1–9 μL. Sequencing-ready libraries are produced in approximately 6 hours with around 1.5 hours of hands-on time, and the panels are compatible with Illumina, Ion Torrent, and Element Biosciences platforms. QIAGEN reports greater than 90% sensitivity for detecting small variants down to 0.5%. Enzymatic cleanups replace bead-based steps, supporting automation-friendly workflows.

When run on the Beckman Coulter Biomek i7 Hybrid workstation, the automated method can process 1–96 samples per run, with target enrichment PCR cycles adjustable between 24 and 36 and ethanol drying adjustable from 1 to 20 minutes. In a 24-sample demonstration, libraries were generated in under 7 hours, with read fragments exceeding 2.9 million and over 86% aligning to the target sequence.
