Digital DNA sequencing with molecular barcodes enables confident detection of low-frequency variants. The QIAseq Targeted DNA Panels from QIAGEN provide a complete Sample to Insight solution for targeted DNA sequencing, incorporating molecular barcodes into starting DNA material before amplification to preserve the uniqueness of each molecule and overcome issues of PCR duplicates, false positives and library bias.

Each panel is a one-box, NGS platform-agnostic solution containing all necessary components to construct libraries from enriched genomic targets. Primer design is based on QIAseq Enrichment Technology, enriching each genomic target with one target-specific primer and one universal primer. All primers required for a panel are pooled into an individual primer pool. Platform-specific indexes are supplied in a separate box, enabling multiplexing of up to 384 samples per sequencing run.

The library construction workflow starts with enzymatic fragmentation of isolated DNA, as low as 20 ng, to generate small pieces of dsDNA. IL-N7 adapters, molecular barcodes and sample indexes are then incorporated into the DNA fragments. Targets are enriched by single primer extension using a single gene-specific primer and a universal forward primer. The final step is library amplification and sample indexing using the IL-S5 sample index primer and a universal primer. The kit contains all reagents (except indexes) for targeted DNA sequencing as a fixed panel for 12 samples, covering less than 100 genes.
