Description

QIAGEN's QIAseq Targeted DNA Booster Panels are designed to complement the QIAseq Targeted DNA Panels workflow. According to the manufacturer, the booster panel consists of a pool of up to 100 primers that can be used to enhance the performance of certain primers in any panel (cataloged, extended, or custom), or to extend the contents of an existing custom panel. The primers are delivered as a single pool that can be spiked directly into the existing panel.

The QIAseq Targeted DNA Panels employ digital sequencing by incorporating molecular barcodes into the starting DNA material before amplification takes place, which helps preserve the uniqueness of starting DNA molecules and addresses issues with PCR duplicates, false positives, and library bias. Each panel is an NGS platform-agnostic, one-box solution containing all necessary components to construct libraries from enriched genomic targets. The primer design uses QIAseq Enrichment Technology, in which each genomic target is enriched by one target-specific primer and one universal primer, reducing the amount of primers required. All primers required for a panel are pooled into an individual primer pool to reduce panel handling. The unique buffer and enzyme system has been optimized to achieve high coverage of GC-rich genomic regions, and platform-specific indexes are contained in a separate box to allow multiplexing of up to 384 samples per sequencing run.

The complete workflow from extracted DNA to sequencing-ready libraries can be completed in 9 hours. Sequencing files can be processed through the QIAseq pipeline, a cloud-based data analysis solution that filters, maps and aligns reads, counts unique molecular barcodes associated with targeted genomic regions, and calls variants using a barcode-aware algorithm. Results can then be fed into IVA or QCI for interpretation.
