QIAseq Stranded RNA Lib Kit UDI-D (96)

The QIAseq Stranded RNA Library Kits from QIAGEN enable preparation of stranded whole transcriptome or mRNA-enriched RNA-seq libraries for next-generation sequencing applications on Illumina instruments. The workflow supports RNA fragmentation, reverse transcription, second-strand synthesis with end-repair and A-addition, adapter ligation, and CleanStart PCR enrichment, with QIAseq Beads included for library cleanups between protocol steps. The kit ships with a plate of Y-shaped adapters carrying unique dual indices (UDI) for sample multiplexing.

Library construction is achievable in less than 5 hours. Reverse transcription employs an RNase H- reverse transcriptase combined with random primers, and the workflow does not require Actinomycin D to maintain strand specificity. The strand-specific ligation step establishes strand specificity without additional reagents or laborious steps, and the HiFi PCR Master Mix uses a high-fidelity, hot-start DNA polymerase for amplification irrespective of GC content. By combining multiple UDI Y-adapter plates from different kits, up to 384 samples can be sequenced together per lane on an Illumina NGS instrument. QIAseq FastSelect reagents, sold separately, can be incorporated to rapidly remove up to 99% of unwanted rRNA and/or globin mRNA, including from degraded RNA inputs such as formalin-fixed paraffin-embedded (FFPE) samples.

The QIAseq Stranded RNA Lib Enzyme Kit and QIAseq UDI Y-Adapter Kit components are stored at −30 to −15°C, and the QIAseq Beads are stored at 2–8°C and must not be frozen. The product is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of a disease.
