QIAseq Stranded RNA Lib Kit UDI-C (96)

From QIAGEN, this stranded RNA library preparation kit includes the UDI-C index set and provides the reagents and unique dual-index adapters needed to create strand-specific NGS libraries suitable for multiplexed sequencing on Illumina instruments.

The kit enables one-day library construction from a broad range of RNA inputs. The workflow covers RNA fragmentation, reverse transcription, second-strand synthesis with end-repair and A-addition, adapter ligation, CleanStart PCR enrichment, and library cleanup using the included QIAseq Beads. During reverse transcription, the optimized RT enzyme and buffers do not require Actinomycin D to enhance strand specificity, while a strand-specific ligation step establishes strand specificity without additional reagents or time-consuming steps.

The kit includes a plate of adapters with different barcodes sealed by a pierceable foil that allows use of defined portions of the plate. Multiple kits with different UDI adapter plates can be combined to multiplex up to 384 samples per lane on an Illumina NGS instrument. For rRNA and/or globin mRNA depletion, QIAseq FastSelect reagents are added separately and can remove up to 99% of unwanted rRNA and/or globin mRNA, including from formalin-fixed paraffin-embedded (FFPE) samples, with the full workflow completed in less than 5 hours.
