QIAseq Stranded RNA Lib Kit UDI-A (96)

Designed for next-generation sequencing applications on Illumina instruments, this library preparation kit from QIAGEN supports the construction of stranded whole transcriptome or mRNA-enriched RNA-seq libraries. The kit includes buffers and reagents for RNA fragmentation, reverse transcription, second-strand synthesis combined with end-repair and A-addition, adapter ligation, CleanStart PCR enrichment, and QIAseq Beads for library cleanups. A plate containing unique dual-index (UDI) adapters with different barcodes, sealed with a pierceable foil, is included to enable sample multiplexing.

The workflow can be combined with QIAseq FastSelect reagents (sold separately) for rRNA and/or globin mRNA removal during library preparation. According to the manufacturer handbook, QIAseq FastSelect removes up to 99% of all unwanted rRNA and/or globin mRNA, even when starting with degraded samples such as formalin-fixed paraffin-embedded (FFPE) material. Strand specificity is achieved without the use of reagents such as Actinomycin D, and library amplification is performed using a high-fidelity, hot-start DNA polymerase via the HiFi PCR Master Mix.

For higher multiplexing, up to 384 samples can be combined per sequencing lane by pooling libraries prepared with the UDI-A, UDI-B, UDI-C, and UDI-D Y-adapter plates. The kit is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of disease.
