QIAseq Stranded RNA Lib Kit UDI (24)

From QIAGEN, this stranded RNA library preparation kit is designed for generating whole-transcriptome or mRNA-enriched RNA-seq libraries for next-generation sequencing on Illumina instruments. The kit provides reagents for the complete workflow, including RNA fragmentation, reverse transcription, second-strand synthesis combined with end-repair and A-addition, strand-specific adapter ligation, and CleanStart PCR enrichment, together with QIAseq Beads for library cleanup between steps.

The protocol achieves strand specificity without using actinomycin D or dUTP, relying instead on optimized reverse transcriptase and buffer chemistry to support sensitive detection of low-expression transcripts with high library complexity. The included CleanStart HiFi PCR Mastermix delivers high-fidelity amplification and protects against carry-over PCR contamination. The kit ships with a plate of barcoded adapters with pierceable foil sealing so individual wells can be accessed as needed; each adapter carries a unique 8-nucleotide i5 and i7 index for multiplexing samples.

Libraries can be produced in 4–5 hours, supporting preparation, QC and the start of a sequencing run within a single working day. The system works with high-quality total RNA or mRNA-enriched samples and tolerates low-quality fragmented input, including degraded FFPE RNA. When the mRNA enrichment protocol is used, more than 96% of reads map to protein-coding regions.
