QIAseq Single Cell RNA Lib Kit UDI (384)

From QIAGEN, this library kit uses unique dual indexes to support RNA-seq library construction from single cells and low-input RNA samples for next-generation sequencing on Illumina instruments. The workflow enables uniformly amplifying all transcripts from single cells or enriching specific RNA, fragmenting the amplified cDNA, and generating a PCR-free library. The kit employs multiple-displacement amplification (MDA) with a HiFi polymerase that minimizes the incorporation of amplification errors, along with HiFi and proofreading polymerases. A one-tube library preparation protocol eliminates sample cleanup between steps, reducing starting material loss and cross-contamination risk, and the co-optimized process is designed to yield NGS-ready libraries in one working day.

Reverse transcription using Quantiscript RT Enzyme Mix proceeds for 60 minutes, followed by ligation of cDNAs for 30 minutes and an isothermal cDNA amplification step lasting 120 minutes. Amplified cDNA can be stored long-term at −30 to −15°C, with sufficient material generated for both NGS library preparation and follow-up experiments such as qPCR.

Per the manufacturer, the enzyme kit and UDI Y-adapter kit are shipped for storage at −30 to −15°C, while the included beads should be stored at 2–8°C and never frozen. The product is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of a disease.
