Description

From QIAGEN, this single-cell DNA library preparation kit provides a complete solution for generating whole genome sequencing libraries from isolated single cells or limited quantities of intact genomic DNA. Optimised for low DNA input, it supports Unique Dual Index (UDI) tagging to reduce index cross-talk and enable confident multiplexed sequencing on Illumina-compatible instruments, including the Illumina NovaSeq [S1][S2]. Indicated applications include single-cell genomic library preparation, copy number analysis, targeted sequencing workflows, and the characterisation of aneuploidy and sequence variation in single cells from rare samples [S1][S2].

The kit relies on three core technologies to deliver high-diversity whole genome libraries. After cell lysis of isolated single cells or small clusters, for example those obtained from laser-capture microdissection platforms, Multiple Displacement Amplification (MDA) using the ultra-high fidelity REPLI-g DNA Polymerase amplifies microgram amounts of gDNA, eliminating PCR duplicates, minimising GC-bias and introducing fewer sequencing errors than high-fidelity PCR enzymes [S1]. The amplified DNA is then cleaved into short inserts by QIAGEN's highly random enzymatic FX DNA fragmentation reaction, end-repaired, A-tailed and ligated to QIAseq Unique Dual-Index Y-Adapters, with cleanup performed using the supplied QIAseq Beads [S1]. Because the workflow is entirely PCR-free, libraries avoid amplification-induced errors and duplicates, supporting high library diversity and comprehensive coverage even in regions with high GC-content [S1]. The full cell-to-library procedure requires only 3.5 hours to complete [S1].

Alongside the whole-genome protocol, the kit provides instructions for unbiased whole genome amplification and for target enrichment of small genomes such as viral DNA or mitochondrial genomes [S1]. Up to 384 samples can be processed in a single flow-cell lane, increasing throughput and lowering per-sample sequencing costs [S1].
