Description

QIAGEN's targeted NGS panels enable combined analysis of RNA fusion events, gene expression and single nucleotide variant (SNV) calling from a single sample. Built on QIAseq Enrichment Technology, which uses one gene-specific primer per target, the kits offer flexibility for targeting fusions and dense overlap of SNV regions of interest. All kits are optimized for use with unique dual index (UDIs) adapters for Illumina® sequencing systems, and catalogued panels cover oncology applications including sarcoma, leukemia and lymphoma, with custom panels supporting both gene-level and transcript-based designs.

In benchmarking work, an analysis using 5 ng of control RNA as input successfully detected eighteen clinically relevant RNA gene fusions. For gene expression, a built-in internal reference control supports normalization across a 5-fold range of RNA input. When used to analyze GM12878 RNA samples, the panels achieved 93.4% sensitivity for SNPs (VMF >=5%) within the covered and expressed target region, and they can also detect SNPs in exoRNA where cancer-specific markers often reside.

The workflow starts with isolated total RNA (10 ng or less), which is reverse transcribed into first-strand cDNA followed by robust second-strand synthesis. Adaptor complexes containing unique molecular indices (UMIs) and sample indices are then incorporated into the dsDNA, after which QIAseq Enrichment Technology chemistry enriches the targets of interest. Libraries are finally amplified by fast universal PCR that adds a second index for dual indexing.
