Description

A library normalization kit from QIAGEN is intended for the normalization of next-generation sequencing (NGS) libraries for sequencing on Illumina instruments without prior library quantification. The kit is designed for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of a disease.

The workflow uses modified library amplification primers together with a magnetic bead–based chemistry to bind and release a defined amount of library molecules in a tightly controlled fashion. Normalized libraries are eluted as double-stranded DNA under non-denaturing conditions with a target concentration of approximately 4 nmol/L, enabling balanced library pooling and optimal clustering on Illumina flow cells. After normalization, libraries can be safely stored at −20°C. The chemistry is compatible with virtually all libraries that can be sequenced on Illumina instruments, including QIAGEN and third-party library preparation workflows.

Library modification using the Normalizer Primer Mix is required prior to normalization. For finished libraries with intact P5/P7 ends, modifications can be added in a separate 2-cycle PCR step (Workflow A). For library preparation workflows that involve ligation of full-length indexed adapters, modifications can be applied in-process during library amplification without an additional PCR step (Workflow B). The kit is not compatible with PCR-free library preparation.
