Description

A hybridization-capture-based target enrichment kit from QIAGEN designed for whole exome sequencing on Illumina instruments. The workflow enriches exonic sequences from indexed whole genome libraries, covering the coding regions of approximately 20,000 human genes — about 1–2% of the human genome. Libraries can be generated from enzymatically fragmented genomic DNA, physically sheared genomic DNA, or circulating cell-free DNA, with as little as 200 ng input per library.

Up to 8 indexed whole genome libraries can be combined in a single hybridization capture reaction, with multiplexing support for up to 384 samples per sequencing run. The hybridization step can be completed in as little as 30 minutes, followed by streptavidin bead-based capture and post-capture amplification designed to minimise GC-content bias. The manufacturer reports that the workflow can reduce exome sequencing costs by up to 50% and supports a single-day, automation-compatible sample-to-sequencing process.

The probe set targets Consensus Coding Sequence (CCDS)–annotated protein-coding regions based on the hg38 genome build, using 5'-biotinylated double-stranded 120 nt DNA probes that capture both strands of about 192,000 target regions. Noncoding transcripts, long UTRs, and low confidence exons are excluded, reducing the total target region to 33 Mb. The included One-4-All Blocking Oligos bind Illumina TruSeq-compatible NGS adapters to enhance capture specificity. The kit is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of disease.
