Description

QIAGEN's QIAseq Human Exome Kit is designed for hybridization-capture based enrichment of human exonic sequences from human whole genome NGS libraries prepared for Illumina instruments. The kit employs 5'-biotinylated double-stranded 120 nt DNA probes that target Consensus Coding Sequence (CCDS)-annotated protein-coding regions of the human exome based on the hg38 genome build, with a total target region size of 33 Mb, capturing both strands of approximately 192,000 target regions.

The workflow supports simultaneous hybridization capture from up to 8 samples with as little as 200 ng input per library, with hybridization achievable in as little as 30 minutes. Pre-capture libraries can be generated from enzymatically fragmented genomic DNA, physically sheared genomic DNA, or circulating cell-free DNA using compatible QIAseq library preparation kits. After hybridization, biotinylated probe-target hybrids are immobilized on streptavidin-coated magnetic beads and washed to remove nonspecific targets. The workflow supports multiplexing of up to 384 unique dual-indexed libraries per sequencing run and provides high coverage uniformity with 99% base-level coverage at ≥20x depth and >98% combined sensitivity for SNVs and Indels.

The kit is supplied as two boxes. Box 1, shipped on dry ice and stored at –30 to –15°C, contains the Human Exome Probe Set, One-4-All Blocking Oligos, One-4-All Blocking Solution, Fast Hybridization Solution, Vapor-Lock, Post Hybrid-Capture PCR Mix (2x), Primer Mix Illumina Library Amplification, and RNase-free Water. Box 2, shipped on cool pack at 4°C and stored at 2–8°C, contains Streptavidin Binding Beads, Post Capture Binding Buffer, Wash Buffer A, and Wash Buffer B. The QIAseq Human Exome Kit is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of a disease.
