Removing ribosomal RNA from total RNA prior to library construction is essential for most RNA-seq workflows, and the QIAseq FastSelect RNA Removal Kit from QIAGEN delivers this depletion in a single-tube, probe-based reaction. Packaged as 24 reactions, the kit uses sequence-specific oligonucleotides that hybridize to unwanted transcripts, which are subsequently enzymatically removed so the RNA of interest remains intact for downstream analysis.

Primary applications include rRNA depletion from total RNA inputs ranging from 10 ng to 1 µg, with full compatibility for intact RNA, partially degraded RNA, and FFPE-derived samples. The workflow integrates directly with QIAseq stranded library prep chemistries as well as other Illumina-compatible library construction methods. Single-use reagents are supplied in individual vials to prevent cross-contamination between reactions.

Handling and storage: store the kit at -20 °C, protect from light, and minimize freeze-thaw cycles by aliquoting working stocks. The hybridization and removal steps occur in a single reaction tube, reducing handling time and limiting sample loss. After depletion, the RNA is ready for immediate library construction without intermediate purification.

Typical use cases include transcriptome profiling when poly(A) selection is unsuitable (for example with degraded inputs), strand-specific RNA-seq after rRNA removal, and studies focused on non-coding RNA species. By eliminating more than 95% of rRNA from human, mouse, or rat total RNA, the workflow preserves lncRNA and other regulatory transcripts, making the 24-reaction format a practical choice for small to medium-throughput projects such as pilot studies, method optimization runs, and focused sample cohorts prepared for sequencing on Illumina platforms.
