Ribosomal RNA depletion from human, mouse, and rat samples is achieved in a single tube with the QIAGEN QIAseq FastSelect RNA Lib HMR Kit, which contains enough blocker reagent for 768 RNA library preparations. The FastSelect chemistry uses short, high-affinity oligonucleotides that anneal to rRNA sequences and prevent cDNA synthesis across 28S, 18S, 5.8S, 5S, and the mitochondrial 12S and 16S transcripts, removing the need for poly(A) selection or hybrid capture.

The workflow is compatible with stranded total RNA library protocols, integrating directly into existing reverse transcription reactions and adding roughly 14 minutes of hands-on time. It accepts RNA inputs from 1 ng to 1 µg, including intact, partially degraded, and FFPE-derived material, and tolerates the fragmented input often obtained from formalin-fixed clinical specimens. Reagents are validated on Illumina sequencing platforms and perform reliably with low-quality RNA where ribo-depletion efficiency is critical for usable transcriptome coverage.

Each kit is supplied in vials pre-aliquoted for 768 individual reactions and should be stored at -20°C to preserve blocker activity. The single-tube format minimizes sample loss and handling, and the chemistry has been shown to maintain transcript representation across GC-rich regions while preserving strand specificity, supporting reproducible total RNA-seq data from human, mouse, and rat samples.
