Description

Designed for the rapid removal of globin messenger RNA during stranded NGS library preparation, QIAGEN's QIAseq FastSelect −Globin kit has been tested with whole blood total RNA samples from human, mouse, and rat, where 95–99% removal of globin mRNA is routinely observed. The kit works with 1 ng − 1 µg of total RNA and removes up to 99% of unwanted cytoplasmic and mitochondrial rRNA and/or globin mRNA from human, mouse, rat, and other mammalian RNA libraries in a single step.

The one-step protocol is integrated into the reverse transcription phase, completing globin mRNA removal in just 14 minutes without additional enzymatic cleanups or separate depletion steps. The kit is compatible with total RNA- and mRNA-enriched samples isolated from cells, fresh or frozen tissue, FFPE tissue, whole blood, and serum/plasma samples, including exosomes, and has been validated with stranded RNA library preparation kits from QIAGEN, Illumina, NEB, and Roche. For total RNA blood samples, the −Globin reagent must be combined with the −rRNA HMR reagent, while only the −Globin reagent is needed for mRNA-enriched samples.

All components should be stored at –30 to –15°C in a constant-temperature freezer. The kit is intended for molecular biology applications only and is not intended for the diagnosis, prevention, or treatment of disease. When globin is being removed, 2 additional cycles of CleanStart® Library Amplification are required in the downstream workflow.
